Regulation of Apoptotic Cell Death in U937 Leukemia Cells by Fatty Acids

Abstract
In the course of screening for substances modulating the apoptosis of U937 human leukemia cells induced by etoposide (10 ¥ìM), a fungal strain F90007 producing a high level of modulator was selected. The active substances were purified and identified as palmitic acid, stearic acid and linoleic acid by spectroscopic methods. Palmitic acid (60 ¥ìM) was found to be the most potent apoptosis inducer in the U937 cells after 8hrs of treatment. Among the saturated fatty acids, myristic acid showed the stronggest inhibitory activity for caspase-3 production, a major protease in the apoptosis cascade, with an IC_(50) value of 80 ¥ìM after 8 hrs of treatment. The simultaneous addition of 40-100 ¥ìM myristic acid with etoposide blocked and abrogated both morphological apoptotic changes and internucleosomal DNA fragmentation.
